Would this combo be a good setup?

Would just the speakers be enough or does the amp add a lot?

Some more questions... Does your sub have to be facing the back for it to be heard while riding?

Does drilling holes in the pylon for the speaker wire make it week enough to worry about?

Do radios have certain amounts of power they can put out because I have an old Glastron with the radio that came with it and wonder if it can put out enough sound for the speakers?

mmchar, No the stock stereo won't be enough. You have a couple of options though:

Stick with that combo but get a 300-600W mono-block amp for the sub

Get a 4 channel amp (at least 400W) and bridge the subwoofer over 2 channels. This is probably the cheapest way out but the sub won't be super loud.

I think the best place for the sub in those boats would be under the driver's side dash.
